Oct-4 (Octamer-binding transcription factor 4) is a transcription factor with a central function in embryogenesis. The protein is important for maintaining the pluripotency of embryonic stem cells and primordial germ cells. Oct-4 is also known as Oct-3, OTF3, and POU5F1 (POU domain class 5 transcription factor 1). Oct-4 is a characteristic marker molecule of embryonic stem cells. Many current publications describe that the immunohistochemical staining of this protein is useful to detect metastatic germ cell tumours. Additionally, it was shown that Oct-4 is superior to other markers because of its high specificity and sensitivity. IGCNU (Intratubular germ cell neoplasia, unspecified type), seminomas, embryonal carcinomas, germinomas, and dysgerminomas are consistently positive for Oct-4.
